I want to get into great cardio shape, but I'm in horrible cardio shape right now. The problem started about a year ago when I sprained my ankle, and since, I have only occasionally done cardio. Furthermore, because of my ankle injury, it's hard for me to do high impact exercises like sprints because the impact may lead to another sprained ankle. Bottom line is, when I do cardio, it's seems pretty low impact (about 5 mph on the treadmill), although I do get my hartrate up between 65-70%. But, isn't that for fat burning? Is doing that kind of cardio going to get me into shape?

What do you mean " ..may lead to another sprained ankle" Has it not healed? Do you still feel pain? I ahve sprained my ankles many times and have been able to resume training without any issues till the next time I stepped on someone's foot and rolled my ankle or something similar. You may need to strengthen the joint up with some wobble board exercises and calf raises.
If that is all the running you are able to do then that will have to do, but to get better, you must try to increase the speed/distance slowly at some point.

Calf exercises help prevent sprained ankles? What are wobble exercises?
It doesn't hurt, but it still feels tight and I can tell that, although it should be, as it's been about 9 months, it's not where it once was. So, I'm reluctant to jeopardize apraining it again as it was a pretty severe sprain.
How would you recommend I commence? Is cardio 3x a week for 30 min. good enough?

Also, are there any supplements that will aid in my progress, such as fiber, which is supposed to be good for your heart. Or B vitamins or something?
